MGF (Mechano Growth Factor) is the native 24-amino-acid E-domain peptide from the IGF-1Ec splice variant - the acute mechanical-loading signal that activates satellite cells as the first phase of skeletal muscle repair.
MGF is a splice variant of IGF-1 produced by alternative splicing of the IGF-1 gene. When a cell includes exon 5 of the gene, the reading frame shifts and a unique C-terminal "E-domain" is produced (the "Ec" in IGF-1Ec). That E-domain, which is 24 amino acids long, is the bioactive part that researchers call MGF.
Unlike mature IGF-1, which acts through the classical IGF-1 receptor pathway, the MGF E-domain acts through a completely different mechanism. It does not bind IGF-1R - instead it appears to target cell nuclei directly for intracrine gene regulation, activating quiescent muscle satellite cells from G0 into the proliferative cell cycle. This makes MGF an early-phase repair signal rather than a terminal-differentiation factor.
